Would anyone have any concerns about using the following with SoftRAID to create a RAID 1+0 on a Mac Pro 5,1 running macOS Sierra 10.12.6?

Dual 2.26GHz Quad-Core Xeons
64GB 1066MHz DDR3 ECC SDRAM
2x Inateck USB 3.0 Cards
4x Toshiba DT01ACA300 3TB Hard Drives (which I will also use with SoftRAID)

There should be no problems. If you get any issues, disable the SMART over USB driver in SoftRAID preferences and see if that helps.
