If I have 3 NVME Drives that total 8TB(4,2,2) and a 16TB HDD, is there a way use a raid to still get fast speeds for Video editing and use the 16TB at the failsafe drive?

You have a mismatch there. Not much you can do. You can RAID 0 the two small drives. Perhaps use Carbon Copy Cloner to target the large HDD for a backup volume.

@softraid-support I have 3 drives in the shuttle, 2TB, 4TB, and 2TB and when I go RAID 0, I can only use 6TB?
Any Idea what that issue is?

All partitions in RAID volumes are identical sized. that is why. You cannot efficiently mix drive sizes
