Hi,
I currently have one SSD which I use in external enclosure, which I intend to move into 4 drive soft raid set up. Do I need to backup the data off this drive before I do this?

No, the enclosure a disk is in does not affect what is on the disk.

to confirm - if I use the used SSD and pair it with 3 new SSDs to be part of a raid 5 setup, it will not get wiped?

It will absolutely be wiped, as you need to initialize all 4 disks to create the volume.
